Campagne d’évaluation du stock de palourdes du bassin d’Arcachon. Année 2018 ArchiMer

This report details the results of clam stock assessment survey, which took place between 14 May and 17 June 2018 in Arcachon Bay. Co-organized with CDPMEM33, with financial support from DLAL FEAMP Arcachon Basin Val de l'Eyre, Nouvelle-Aquitaine region, the CDPMEM33 and Ifremer, the clam stock assessment and its characteristics were provided. The samples were collected from 533 sampling stations distributed over an area of 52 km², with an Hamon grab, using a generalized random tessellation stratified spatially balanced design (GRTS). Venerupis philippinarum remains the dominant species with 99 % of the population. The size distribution of Manila clam is in the range 5 – 41 mm with a main mode around 24-28 mm. The Gram-negative bacterium Vibrio tapetis is known as the causative agent of Brown Ring Disease (BRD) in the Manila clam Venerupis (=Ruditapes) philippinarum. This bivalve is the second most important species produced in aquaculture and has a high commercial value. In spite of the development of several molecular methods, no survey has been yet achieved to rapidly quantify the bacterium in the clam. Immune responses of phenoloxidase and superoxide dismutase in the manila clam Venerupis philippinarum challenged with Vibrio tapetis – Part I: Spatio-temporal evolution of enzymes’ activities post-infection ArchiMer

Manila clams, Venerupis philippinarum (Adams and Reeve, 1850), were experimentally challenged with two Vibrio tapetis strains: CECT4600T, the causative agent of Brown Ring Disease (BRD); and LP2 supposedly non-pathogenic in V. philippinarum. Changes in phenoloxidase (PO) and superoxide dismutase (SOD), two major enzymes involved in immunity, were studied in two tissues, the mantle and hemolymph for 30 days after infection in the extrapallial cavity. Bacterial infection in V. philippinarum resulted in modulation of PO and SOD activities that was both tissue- and time-dependent. For species' dynamics highly influenced by environmental conditions, sustainable management for conservation and/or exploitation purposes requires the ability to segregate natural processes from anthropogenic impacts. In this context, identification of phenotypic variations in morphological patterns, and clarification of the mechanisms involved, constitute useful tools to identify adequate management measures from Europe-wide to local conditions. Optimization of a survey using spatially balanced sampling: a single-year application of clam monitoring in the Arcachon Bay (SW France) ArchiMer

Bivalves are important components of benthic marine and freshwater ecosystems throughout the world. One of the most exploited bivalves used for human consumption is manila clam (Venerupis philippinarum). In Arcachon Bay (SW France), commercial fishers and scientists have developed a monitoring survey to estimate clam stocks to assist in implementing a sustainable management strategy. The survey design that is currently used is based on standard stratified random sampling (StRS). The survey has been undertaken every 2 years since 2006. Manila clam stock from Arcachon Bay, France, is declining, as is commercial harvest. To understand the role of environmental biotic interactions in this decrease, effects of a toxic dinoflagellate, Alexandrium ostenfeldii, which blooms regularly in Arcachon bay, and the interaction with perkinsosis on clam physiology were investigated. Vibrio tapetis Displays an Original Type IV Secretion System in Strains Pathogenic for Bivalve Molluscs ArchiMer

The Brown Ring Disease (BRD) caused high mortality rates since 1986 in the Manila clam Venerupis philippinarum introduced and cultured in Western Europe from the 1970s. The causative agent of BRD is a Gram-Negative bacterium, Vibrio tapetis, which is also pathogenic to fish. Here we report the first assembly of the complete genome of V. tapetis CECT4600T, together with the genome sequences of 16 additional strains isolated across a broad host and geographic range. Our extensive genome dataset allowed us to describe the pathogen pan-and core genomes and to identify putative virulence factors.
